Conduct research on ribosomes in yeast, planning, conducting and evaluating results of experiments for broader research on the ribosome associated complex. Focus experiments on specifically identifying the binding sites of Zuo1 to the ribosomal RNA. Utilise RNA bindR software to predict binding sites in the protein sequence, preparing mutants with a single aa exchange for Linker binding. Conduct PCR, cloning and E. coli transformation.Introduced the same mutations in yeast and examine for viability.

Strategised, conducted and evaluated experiments to identify likely new proteins in the 3’ end processing cleavage complex in mammals. Attempted to assemble the complex on immobilized, radioactively labeled mRNA. Carried out experiments after preparing nuclear cell extracts from 10L HeLa cell cultures. Used SDS-PAGE, silver staining, Western blot and autoradiography in conducting analyses, and molecular biology techniques to prepare different mRNA constructs. Checked for quality throughout the process.

Contributed scientific expertise, working to identify probable new proteins. Worked independently to reconstitute the complex with purified proteins derived from human cell lines. Prepared DNA plasmids coding with a HisFlag tag for numerous known protein subunits and conducted transient transfection of HEK and HeLa human adherent cells. Isolated stable cell lines, expressing proteins and monitored transfection with GFP. Used depleted nuclear extract assays, MS-MS, Western blot and silver-stained gels.
