Role of the RNA binding proteins (PPR containing) in the translation complex of kinetoplasts of Trypanosoma brucei. Main achievements: I proved the relevance of the plant PPR-RNA recognition code in the Trypanosoma brucei system. Main techniques: genes knockout in cell lines by homologous recombination; implementation of CRISPR/CAS9 genome editing system for genes substitution; CLIP/CLAP RNA protection studies.

Ulm University, Institute for Molecular Botany
Investigation of the RNA editing machinery in plant organelles Main achievements: I separate the editing activity from the mitochondrial protein extract with subsequent purification of the editing complex subunits using RNA target as a bait. Main techniques: mitochondria and chloroplast isolation; protein overexpression and purification; in vitro RNA-protein and protein-protein interaction studies; CoIP (co-immune precipitation); RIP-CHIP (RNA immune precipitation), FPLS, HPLS.

Mapping a genes coding plant RNA editing factors. Main achievements: I mapped and annotated several PPR (PentatricoPeptide Repeat) proteins as editing recognition factors, such as MEF (mitochondrial editing factor) 3, MEF11, MEF14, MEF13, MEF8, MEF8s. Main techniques: clone based gene mapping; protoplasts isolation and transfection; agrobacterial plant transformation.

Recognition of a cluster of RNA editing sites in the atp4 mRNA Main achievements: The mode of the recognition of clustered sites was analysed. Increase of in vitro RNA editing reaction efficient from 1-5% to 80-95%, which open the opportunity for purification of editing complex. Main techniques: mitochondrial proteins purification; in vitro RNA editing reaction; RT-PCR, DNA sequencing.
