Arbeiten an der Charité

The Giesecke-Thiel Lab investigates initiation, reactivation, and lifestyle of human B cell memory. This includes studying the immunological imprinting of human memory B cells from different antigen exposures, including both the nature of antigen and the anatomical site of priming. Our goal is to identify fundamental mechanisms of human immune memory and to leverage these discoveries to develop new ways to detect, prevent, and modify infection courses and outcomes.

Die Stelle im Überblick

In this project, the successful candidate will explore the phenotypic and molecular features of antigen-specific human memory B cells (mBCs) shaped by mucosal priming in comparison to systemic vaccinations. The candidate will optimize flow cytometry–based antigen-specific B cell stainings and phenotyping strategies and perform antigen-specific B cell stainings, flow cytometry analyses, and cell isolations using FACS and MACS to obtain high-quality antigen-specific populations.
A core responsibility includes establishing, benchmarking, and applying single-cell ATAC- and RNA-sequencing workflows (FLASH-seq), carried out in close collaboration with the local Core Facility. The candidate will additionally carry out basic bioinformatic analyses, including quality control, preprocessing, dimensionality reduction, and exploratory interpretation of single-cell multiomic datasets, with support from collaborating bioinformaticians.

A key task is to investigate how antigen challenges and rechallenges affect human memory B cells, thereby advancing our understanding of immune memory (re-)programming or its resilience to perturbation.

Danach suchen wir

  • Solid background in immunology or molecular cell biology, with a strong interest in human B cell biology and immune memory
  • Hands-on experience with flow cytometry, including multicolor panel design, data acquisition, and analysis
  • Experience in cell isolation techniques, including primary human material, FACS and/or MACS, or willingness to acquire advanced sorting skills
  • Basic practical or conceptual experience with single-cell sequencing technologies (scRNA-seq and/or scATAC-seq), or a strong interest in learning and applying these methods
  • Ability to perform basic bioinformatic analyses of single-cell datasets, including quality control, preprocessing, and exploratory data analysis (e.g. R or Python), or strong willingness to acquire these skills
  • Intellectual curiosity with a strong sense of accountability
  • Strong organizational skills
  • Strong communication skills
  • Ability to work independently as well as collaboratively in an interdisciplinary environment involving wet-lab scientists, core facilities, and bioinformaticians
  • Very good written and spoken English communication skills
