Development and optimization of multiplex PCR assays for the detection and discrimination/typing of viral and/or bacterial pathogens

The focus of my work was the development and optimization of a novel immuno assay (Proximity Ligation Assay; PLA) in close collaboration with Applied Biosystems. PLA is an ultra-sensitive method for the detection of proteins, protein interactions and protein modifications which is based on the proximity-dependent ligation of antibody-coupled oligonucleotides. The ligation products finally serve as templates for the read-out and quantification via real-time-PCR.

The focus of my PhD thesis was to develop new target and signal amplification techniques for expression profiling on oligonucleotide microarrays, using minimal amounts of starting material.
