My PhD focused on how cells accurately divide their replicated genetic material equally between the two rising daughter cells during mitosis. Specifically, I investigated the role of the Chromosomal Passenger Complex in facilitating chromosome bi-orientation. To achieve that goal, we applied a combination of in vitro biochemical reconstitution and cell-based functional assays.

Independently designed and conducted CRISPR/Cas9 mediated genome editing. This work included largely standard molecular biology and tissue culture techniques, i.e. cloning (primer design, PCR), sequencing, cell transfection (lipid-based & electroporation), screening cell lines (genomic DNA PCR, Western Blot).

During this voluntary internship I learned how to design PCR- and sequencing-based strategies to diagnose genetic disorders. Additionally, I gained working experience in a GMP-certified lab and helped to prepare an Audit.
