UK NEQAS for Leucocyte Immunophenotyping
The majority of being a Medical Technical Officer for the molecular genetics side of UK NEQAS LI entails the planning, production of and quality assessment of haemato-oncological EQA samples. UK NEQAS LI EQA samples can consist of mutation-carrying and/or negative cell lines cultured in house, blood products, samples from consenting patients and whole genome amplified DNA. These samples are manufactured to represent typical diagnostic samples for the programmes that UK NEQAS LI offer EQA for.

When working within the genotyping team within the molecular genetics section of this NHS diagnostic lab, I recieved clinical patient DNA/cDNA from the extraction team and processed it using the appropriate molecular genetic assay. When working within the cytogenetics section, I recieved clinical patient metaphase chromosomes fixed on slides from the pre-processing team and analysed them using the appropriate genetic assay.

DNA extraction (from PB, BM, Ficoll isolated WBC, saliva, buccal swabs, solid tumours, tissue, FFPE, guthrie cards, chorionic villi and amniotic fluid) via automated robots (Chemagen 360/EZ1) or manual techniques (Ficoll/QiaAMP DNA kit) to provide decent yields of quality nucleic material for downstream molecular diagnostics assays. Adept at manual TRIzol RNA extraction from lyophilised cells, PB or BM followed by cDNA synthesis using the ThermoFisher Superscript IV VILO kit in a reverse-transcription PCR.
