My position as a Postdoctoral Research Fellow in the laboratory of Dr. Yi Feng focuses on the establishment of a new skin specific cancer model to study the earliest events of preneoplastic cell progression and innate immune cell interaction using the zebrafish, Danio rerio. By use of this model I aim for translational profiling of individual cell lineages of zebrafish skin tissue to study gene expression profiles of preneoplastic keratinocytes.
I was investigating the contribution of extracellular matrix to embryonic fin development and cutaneous wound healing using the zebrafish, Danio rerio. One of my PhD projects included the first description of cutaneous wound closure in adult zebrafish. Therefore an adult wound closure assay was established, which preserves the scale-skin architecture. The establishment of a live imaging technique for adult zebrafish revealed rapid wound re-epithelialization in this new model system for wound closure.
The work focused on the adaptation of the DHFR heat-inducible degron for protein inactivation, which we successfully utilized for the first time in a multicellular organism: Arabidopsis thaliana. My work provided me with excellent training with this model organism that included transgenesis based on the use of the Agrobacterium tumefaciens, delicate heat shock treatments, and western blot analysis.
